KERAGAMAN BAKTERI YANG BERPOTENSI SEBAGAI AGEN PENDEGRADASI LIMBAH MASKER BERBAHAN POLIPROPILEN YANG DIISOLASI DARI PANTAI PARANGTRITIS DAN TANJUNG PASIR

Abstract


Penelitian ini bertujuan untuk mengetahui karakteristik, jenis bakteri yang dapat mendegradasi limbah masker nonmedis duckbill hitam berbahan polipropilen, serta aktivitas degradasi polipropilen oleh 10 isolat bakteri pada limbah masker yang ditemukan di pesisir Pantai Parangtritis, DI Yogyakarta dan Tanjung Pasir, Banten. Penelitian dilakukan dengan pengukuran nilai optical density isolat bakteri, uji degradasi zona bening, karakterisasi secara morfologi, fisiologi, serta profile matching dengan karakter bakteri acuan sesuai Bergeys’s Manual of Determinative Bacteriology dan referensi ilmiah pendukung untuk menentukan dugaan genus isolat. Hasil karakterisasi fenotipik dikonstrusikan dengan dendogram untuk mengetahui indeks similaritas. Hasil penelitian diketahui bahwa kemampuan isolat bakteri limbah masker mendegradasi polipropilen dalam kategori sedang (diamater 1,0 – 2,5 mm). Karakter isolat yaitu bakteri gram positif dan negatif; berbentuk coccus dan basil; bakteri dominan tidak membentuk endospora, bentuk semua koloni circular; elevasi flat dan raised; margin undulate, entire, dan lobate; warna putih susu dan putih kekuningan; dan hasil uji biokimia beragam. Hasil dendogram didapatkan 3 jenis (genus) bakteri yaitu Pseudomonas sp., Rhodococcus sp. dan Halomonas sp.
